Abstract

 

Blood storage plays a crucial role in maintaining blood availability for transfusion, but it can impact the quality of blood components, including hemoglobin levels. This study aimed to determine the differences in hemoglobin levels in whole blood components before and after 5 days of storage at the Indonesian Red Cross (PMI) UTD in Medan City. The study used an observational analytical design with a quantitative approach to 41 whole blood samples selected using a simple random sampling technique. Hemoglobin levels were measured using a Sysmex XP-300 Hematology Analyzer. Data analysis was performed using the Shapiro–Wilk normality test and paired sample t-test. The results showed that the average hemoglobin level increased from 13.422 g/dL before storage to 14.598 g/dL after storage. The paired sample t-test showed a p-value <0.001, indicating a significant difference between hemoglobin levels before and after storage. The 8.76% increase in hemoglobin levels is suspected to be related to erythrocyte hemolysis during storage. It was concluded that 5 days of whole blood storage causes an increase in hemoglobin levels, so storage time and conditions need to be considered to maintain blood quality and the accuracy of laboratory test results.
